The Nucleic Acid Shared Resource operates two Transgenomic (www.transgenomic.com) Wave DHPLC systems (denaturing high-performance liquid chromatography) to support the following: Scan DNA fragments for both known and novel mutations and/or SNPs without extensive resequencing or complex comparative DNA sequence analysis. Detect and/or enrich for low-abundance mutations in heterogenous clinical specimens that contain coexisting tumor and normal tissue. Fractionate complex mixtures of related DNA fragments based on differences in DNA sequence content; applications include molecular analysis of mixed microbial populations. Perform various related molecular assays, including primer-extension genotyping and analysis of end-point products from various PCR or RT-PCR protocols. Please submit 25 µl unlabeled PCR products in a PCR plate or 0.2 ml strip tubes for mutation analysis. No purification is necessary. Submit the sequence of the PCR products including both primers by e-mail and indicate the position of the polymorphism. After the run we create PDF files of the colored chromatograms.
