The Nucleic Acid Shared Resource operates two Transgenomic (www.transgenomic.com) Wave DHPLC systems (denaturing high-performance liquid chromatography) to support the following:
Scan DNA fragments for both known and novel mutations and/or SNPs without extensive resequencing or complex comparative DNA sequence analysis.
Detect and/or enrich for low-abundance mutations in heterogenous clinical specimens that contain coexisting tumor and normal tissue.
Fractionate complex mixtures of related DNA fragments based on differences in DNA sequence content; applications include molecular analysis of mixed microbial populations.
Perform various related molecular assays, including primer-extension genotyping and analysis of end-point products from various PCR or RT-PCR protocols.
Please submit 25 µl unlabeled PCR products in a PCR plate or 0.2 ml strip tubes for mutation analysis. No purification is necessary. Submit the sequence of the PCR products including both primers by e-mail and indicate the position of the polymorphism. After the run we create PDF files of the colored chromatograms. We provide support for the evaluation for detection of heterozygotes at no extra charge.
